Tien-Chen Lin

Topic
From oscillating cytoskeletons to stable neuronal polarity
Abstract
Neurons acquire polarity by selecting one neurite as the axon while the others become dendrites, yet how this asymmetry is established remains unclear. Using live imaging and genetic loss-of-function approaches in vivo, together with optogenetic control and local cytoskeletal perturbations in cultured neurons, we identified a soma-initiated oscillatory program that coordinates axon selection. Periodic ARP2/3-dependent actin branching remodels a global actomyosin network, generating recurrent cycles of neurite extension and retraction that transiently bias individual neurites toward axon fate. As polarization proceeds, one neurite escapes this oscillatory regime and enters persistent growth, while actomyosin activity continues to constrain competing neurites. These findings recast neuronal polarization as a dynamic whole-cell process. I will discuss how this cytoskeletal oscillator establishes neuronal polarity and how transient differences in growth may subsequently be converted into stable axonal and dendritic identities.
Biography
Tien-Chen Lin is a postdoctoral researcher in Frank Bradke’s laboratory at the German Center for Neurodegenerative Diseases (DZNE) in Bonn. He trained in biochemistry and microtubule biology at the University of Heidelberg, where he studied the regulation and assembly of microtubule-nucleation machinery. At DZNE, he shifted to neuronal cell biology and led a 2026 Nature study identifying a soma-initiated ARP2/3–actomyosin oscillatory program that establishes neuronal polarity. His current work investigates how transient cytoskeletal dynamics are converted into stable neuronal compartment identities using quantitative imaging, spatial perturbation, mechanistic biochemistry and mathematical modeling.
